Evommune Drops EVO756 for Atopic Dermatitis After Phase 2b Failure

Evommune is discontinuing the development of its oral drug EVO756 for moderate-to-severe atopic dermatitis after the therapy missed its primary and secondary endpoints in a Phase 2b clinical trial. The setback prompts a strategic pipeline shift for the biotechnology firm, which will now redirect the drug toward migraine prophylaxis while advancing a separate candidate for skin conditions.

"EVO756 was generally well tolerated with a safety profile consistent with prior studies, supporting its continued evaluation in migraine prophylaxis," said Dr. Eugene Bauer, Chief Medical Officer of Evommune, in a statement regarding the trial outcome.

Clinical Trial Design and Results

The randomized, double-blind, placebo-controlled trial enrolled 121 adult participants to evaluate multiple doses of EVO756 over a 12-week treatment period, followed by safety monitoring through Week 14. According to an Investing.com report, the compound failed to achieve statistical significance across all tested dose levels for both primary and secondary endpoints.

Phase 2b trials are designed to establish optimal dosing and assess preliminary efficacy in patient populations before companies commit to larger, more expensive Phase 3 pivotal programs. The failure at this stage eliminates atopic dermatitis as an indication for EVO756, though safety data gathered during the 12-week study will inform its ongoing evaluation in neurological applications.

Pipeline Pivot and Financial Runway

With EVO756 removed from its dermatological pipeline, Evommune plans to focus its atopic dermatitis efforts on EVO301, an interleukin-18 binding protein fusion protein. The company intends to initiate a Phase 2b clinical trial for EVO301 by mid-2027.

Despite the clinical setback in its lead dermatology program, executive leadership indicated that the firm retains sufficient capital to fund ongoing research and development.

"The company has a balance sheet supporting operations into 2029," said Luis Peña, President and Chief Executive Officer at Evommune, in a statement.
