Citizens Keeps ProQR $8 Target on Bile Acid Data

Citizens maintains its Market Outperform rating and $8.00 price target on ProQR Therapeutics following a biliary atresia investor event.

Citizens reiterated its Market Outperform rating and $8.00 price target on ProQR Therapeutics N.V. (NASDAQ:PRQR), citing upcoming clinical catalysts for liver therapy candidates AX-0810 and AX-0811 following a recent investor event.

"We believe the current share price does not reflect derisked data generated to date for AX-0810," Citizens said in a note following a presentation by pediatric hepatologist Dr. Gideon Hirschfield.

Clinical Timelines for AX-0810 and AX-0811

ProQR is advancing two primary hepatic candidates through its development pipeline. According to the Investing.com report, the company plans to release additional data for AX-0810 by year-end, followed by first-in-human data for the sodium taurocholate cotransport polypeptide (NTCP) modulator AX-0811 in early January.

Management anticipates that AX-0811 will serve as the lead candidate for subsequent development stages. The upcoming readouts arrive as the company works to close a valuation gap with broader biotechnology benchmarks.

Interim Trial Data and Stock Performance

ProQR shares have declined 18% year-to-date, trailing the SPDR S&P Biotech ETF (XBI) index, which has posted a 29% gain over the same period. Citizens noted that this underperformance contrasts with supportive early-stage clinical metrics.

Interim data for AX-0810 demonstrated several key pharmacological properties:

  • An up to 8-fold dose-dependent increase in serum bile acid levels
  • Favorable shifts in conjugated bile acid and tauroursodeoxycholic acid (TUDCA) profiles
  • An absence of serious adverse events or pruritus during evaluation
  • An extended eight-week half-life supporting infrequent dosing intervals

Understanding Biliary Atresia and NTCP Modulation

Biliary atresia is a rare pediatric liver disease characterized by the progressive inflammation and obliteration of extrahepatic bile ducts, leading to bile duct obstruction, rapid liver fibrosis, and end-stage liver disease if left untreated. The condition typically requires surgical intervention such as the Kasai procedure in infancy, but many patients ultimately require liver transplantation.

Therapeutic candidates like AX-0810 and AX-0811 target bile acid transport mechanisms to mitigate hepatic injury. NTCP modulators specifically regulate the uptake of bile acids into hepatocytes, offering a pharmacological approach to managing cholestatic liver diseases where traditional surgical options are insufficient or need adjunctive medical management.
