Barclays Downgrades Scholar Rock to Equalweight on Valuation
Barclays downgraded Scholar Rock to Equalweight on valuation following Isembyld's approval, while raising its price target on higher drug pricing.

Barclays downgraded Scholar Rock (NASDAQ:SRRK) to Equalweight from Overweight, pointing to a more balanced risk-reward profile following the commercial approval of its lead drug.
At the same time, the firm lifted its price target on the biotechnology company to $58 from $55, driven by higher-than-expected annual pricing for the newly approved treatment.
FDA Approval and Commercial Launch
The rating change follows the U.S. Food and Drug Administration's approval of apitegromab, marketed as Isembyld, as a muscle-targeted therapy for spinal muscular atrophy. The regulatory greenlight covers both adult and pediatric patients aged two years and older who are currently receiving a survival motor neuron, or SMN-targeted, therapy.
Spinal muscular atrophy is a rare genetic neuromuscular disease characterized by the progressive loss of motor neurons and muscle weakness. While existing SMN-targeted therapies focus on addressing the genetic root cause by increasing production of the SMN protein, Isembyld is designed to protect and improve muscle function directly.
Pricing Dynamics and Valuation Assumptions
The upward revision in Barclays' price target stems directly from Scholar Rock's commercial pricing strategy for the drug. The company announced a net annual price of $310,000 for Isembyld, according to analyst coverage details.
That figure outpaces previous modeling assumptions used by Wall Street. Barclays had previously estimated an annual net price of $250,000 for the therapy. The $60,000 variance per patient significantly alters long-term revenue projections, prompting analysts to adjust their valuation models upward despite moving to a neutral rating.
Analyst Outlook on Risk-Reward
While higher pricing strengthens the revenue outlook, the transition from a clinical-stage developer to a commercial-stage enterprise alters the equity's risk profile. Analyst downgrades following successful product approvals are common in the biotechnology sector as much of the near-term regulatory risk is removed and execution risk takes center stage.
With the stock having appreciated around the approval milestone, the adjustment to an Equalweight rating indicates that the shares are viewed as fairly valued at current trading levels relative to the newly priced commercial opportunity.
