Seaport Therapeutics shares rose 5.5% after presenting Phase 1 clinical data for GlyphAgo at the 39th European College of Neuropsychopharmacology Congress in Munich, Germany, showing improved bioavailability for its anxiety treatment candidate compared to traditional agomelatine.

“The data presented at ECNP provide important clinical validation that our Glyph™ platform can meaningfully improve the pharmacokinetic profile of medicines historically limited by high first-pass liver metabolism,” said Daniel Bonner, Ph.D., Co-Founder and Senior Vice President, Platform, at Seaport Therapeutics.

Phase 1 Clinical Trial Results and Pharmacokinetics

The Phase 1 trial evaluated GlyphAgo in 174 healthy volunteers, according to an Investing.com report. The results demonstrated a 6.8-fold increase in bioavailability of agomelatine compared to unmodified agomelatine.

Additionally, the trial recorded 10-fold lower pharmacokinetic variability among participants. These metrics address historical limitations of agomelatine, a medication often constrained by rapid breakdown in the liver before reaching systemic circulation.

Clinical Development Timelines for GlyphAgo

Following the completion of the Phase 1 evaluation, Seaport has initiated a Phase 2a clinical trial. This mid-stage study is evaluating GlyphAgo in adults diagnosed with generalized anxiety disorder and accompanying sleep disturbance, with topline data expected in early 2028.

Alongside the ongoing Phase 2a study, the company outlined future development milestones. Seaport plans to launch a broader Phase 2/3 trial in the first half of 2027 to further assess the drug candidate's efficacy and safety profile in larger patient populations.

Market Response and Stock Valuation

In the public markets, Seaport Therapeutics Inc (NASDAQ: SPTX) traded at $17.60, representing a 5.52% increase from its previous close of $16.68, according to Finnhub market data.

The valuation movement follows investor reception of the pharmacokinetic data presented in Munich. Drug developers utilizing proprietary delivery platforms like Glyph aim to overcome metabolic hurdles that have previously restricted central nervous system therapeutics.

Overcoming First-Pass Metabolism in Drug Design

First-pass metabolism occurs when the concentration of a drug is significantly reduced before it reaches the systemic circulation, typically due to liver enzymes breaking down the active pharmaceutical ingredient after oral ingestion.

By chemically modifying molecules to interact with natural lipid absorption pathways, platforms such as Seaport's Glyph technology attempt to bypass standard hepatic degradation. This mechanism aims to deliver more consistent drug exposure to the target tissues while reducing the high inter-patient variability often observed with unmodified compounds.
