Rhythm Pharmaceuticals has officially launched IMCIVREE (setmelanotide) in Japan to treat acquired hypothalamic obesity (HO). This is the first therapy approved for the condition in the country. The drug's entry into the Japanese market follows its inclusion on the National Health Insurance drug price list. This provides a treatment pathway for a patient population that previously lacked dedicated medical options.
"This launch marks an important advancement for people living with acquired HO in Japan where there were no approved therapies for this disease until now," said Yann Mazabraud, Executive Vice President and Head of International at Rhythm, in a statement regarding the release.
Clinical Efficacy and Trial Data
The regulatory approval in Japan was supported by data from the Phase 3 TRANSCEND trial. According to the company's announcement, the study demonstrated an 18.8% placebo-adjusted reduction in body mass index (BMI) among participants.
IMCIVREE functions as a melanocortin-4 receptor (MC4R) agonist. By activating this pathway, the drug regulates appetite and energy expenditure in patients whose hypothalamic function has been impaired. This impairment often occurs due to tumors, surgical procedures, or radiation therapy. These conditions typically lead to severe, uncontrollable hunger and rapid weight gain.
Market Scope and Patient Impact
Rhythm Pharmaceuticals estimates that approximately 5,000 to 8,000 people are living with acquired hypothalamic obesity in Japan. Because the condition is secondary to other medical interventions, these patients have historically faced limited management strategies beyond intensive lifestyle modifications. These modifications frequently prove insufficient for those with hypothalamic damage.
By securing a position on the National Health Insurance drug price list, the company has established a formal reimbursement framework. This allows patients access to the therapy within the Japanese healthcare system. This launch is part of Rhythm's international expansion strategy as the firm seeks to address rare genetic and acquired forms of obesity in key global markets.
