Guggenheim lowered its price target on Ionis Pharmaceuticals to $80 from $89 while maintaining a Buy rating, citing revised sales projections for key drug assets and recent clinical trial setbacks.
"Guggenheim updated its model to eliminate pelacarsen following the failure of the Lp(a)-HORIZON trial," according to an Investing.com report.
Physician Polls and Tryngolza Projections
The downward revision stems in part from updated modeling for Tryngolza. To gauge future adoption rates, Guggenheim conducted a poll surveying 30 U.S. cardiologists and lipidologists regarding commercial prospects.
Survey respondents projected approximately 40% year-end 2027 adoption of Tryngolza among all severe hypertriglyceridemia patients. Based on these estimates and broader market conditions, Guggenheim models peak sales of approximately $2.9 billion for Tryngolza, sitting just below management guidance of more than $3 billion.
Pipeline Adjustments and Zanvastro Expectations
Alongside the adjustments to Tryngolza, the Guggenheim analyst note stripped out prior contributions from pelacarsen. The removal follows the unsuccessful outcome of the Lp(a)-HORIZON trial, which forced a re-evaluation of Ionis Pharmaceuticals (NASDAQ:IONS) long-term cardiovascular pipeline.
Offsetting some of those reductions, Guggenheim incorporated commercial expectations for Zanvastro following its recent regulatory approval for Alexander Disease. Analysts at the firm model peak sales of approximately $160 million for Zanvastro.
What Is at Stake for Ionis Shareholders
The recalibration of analyst models highlights the balance between commercial execution and clinical trial risk for biotechnology firms. With pelacarsen removed from forward estimates, the valuation of Ionis relies heavily on the successful market penetration of approved treatments like Tryngolza and Zanvastro as the company works to meet its multi-billion-dollar revenue targets.
