Edgewise Therapeutics has initiated patient dosing in its Phase 2 EQUINOX-HFpEF clinical trial evaluating EDG-15400 for adults with symptomatic heart failure with preserved ejection fraction, according to a PR Newswire release. The trial launch coincides with shares of Edgewise Therapeutics Inc (EWTX) trading down 5.64% at $36.28, compared to a previous close of $38.45, according to Finnhub market data.
"Dosing the first patient in the EQUINOX-HFpEF trial marks an important milestone in evaluating EDG-15400's potential to improve the impaired cardiac relaxation that characterizes HFpEF," said Kevin Koch, Ph.D., President and Chief Executive Officer of Edgewise Therapeutics.
Trial Parameters and Patient Demographics
The multicenter, randomized, double-blind, placebo-controlled study is designed to enroll approximately 90 adults. Eligible participants range in age from 30 to 84 years and must have a left ventricular ejection fraction of at least 50%, alongside New York Heart Association functional class II or III symptoms.
Patients enrolled in the study will receive once-daily oral doses of either 25 mg or 50 mg of EDG-15400, or a matching placebo, over a treatment period of up to 12 weeks.
"HFpEF remains one of the most challenging forms of heart failure to treat, in part because current therapies do not directly address impaired cardiac relaxation," said James L. Januzzi, M.D., Chair of the EQUINOX-HFpEF Executive Committee and the Hutter Family Professor of Medicine at Harvard Medical School.
Understanding Heart Failure with Preserved Ejection Fraction
Heart failure with preserved ejection fraction occurs when the heart muscle contracts normally during the pumping phase but fails to relax adequately between beats. This stiffness prevents the left ventricle from filling properly with blood, leading to elevated pressures and fluid buildup in the lungs.
Traditional heart failure treatments often focus on managing fluid retention or reducing blood pressure, while experimental compounds like EDG-15400 aim to target the underlying biomechanical mechanisms of cardiac relaxation. The Phase 2 EQUINOX-HFpEF study is structured to assess whether direct intervention can safely alter this trajectory in symptomatic patients.
