Camp4 Advances SYNGAP1 Trial Design After Cantor Update

Camp4 Therapeutics is advancing the clinical development pipeline for its lead SYNGAP1 program, utilizing regulatory clearances across international jurisdictions as U.S. review continues. At the 12th Annual Cantor Fitzgerald Global Healthcare Conference, Camp4 Therapeutics executives detailed trial designs for its regRNA platform during steady market trading.
"Every 3 to 5 years, you learn about a new rare disease that causes you to say, 'I've never heard of that,' and then it becomes a very important rare disease," said Chief Executive Josh Mandel-Brehm at the conference regarding SYNGAP1 awareness.
Global Regulatory Approvals and FDA Status
Camp4 has secured approvals to start its Phase I/IIa multiple ascending dose (MAD) study for its SYNGAP1 program in Australia, Argentina, and the United Kingdom, according to the Cantor conference transcript. Meanwhile, review by the U.S. Food and Drug Administration remains in progress.
The planned trial is set to enroll about 30 subjects across three dose cohorts. The study utilizes a placebo-controlled design with crossover options, beginning with older sentinel patients aged 8 to 10 before moving down to younger children aged 2 to 3.
"We think it's important to have a control," Chief Executive Josh Mandel-Brehm said, "Namely because if we see great results, we want to have as much optionality as possible to have robust discussions about how to quickly advance this."
Understanding regRNA Platforms and SYNGAP1
SYNGAP1 haploinsufficiency is a rare genetic disorder caused by mutations in the SYNGAP1 gene, leading to intellectual disability, epilepsy, and other neurological impairments. Camp4's regRNA platform aims to target regulatory RNAs to upregulate gene expression, attempting to restore normal protein levels in conditions where one functional copy of a gene is insufficient.
