Lexicon Sets Q1 2027 Readout for Sotagliflozin HCM Trial

Lexicon Pharmaceuticals outlined its clinical pipeline and upcoming regulatory catalysts at the Citi Biopharma Back to School Summit, pointing to a first-quarter 2027 readout for its lead heart drug sotagliflozin as the company's defining upcoming milestone, according to a transcript from Investing.com.
The upcoming data release centers on the Phase 3 SONATA-HCM trial, which evaluates sotagliflozin in patients with hypertrophic cardiomyopathy, a genetic heart condition that causes the thickening of heart muscle walls and restricts blood flow.
"That's the North Star," executives said regarding the upcoming Q1 2027 data release from the SONATA-HCM Phase 3 trial during the Citi summit presentation, highlighting its central role in the company's clinical valuation.
Preparing for Type 1 Diabetes Resubmission
Beyond the hypertrophic cardiomyopathy program, Lexicon is advancing preparations to resubmit its regulatory application for sotagliflozin in type 1 diabetes. The company completed necessary patient exposure tracking at the end of August and is actively compiling the final data package for review.
"We achieved that exposure at the end of August. The data is done. Now we just are in the throes of preparing it and getting ready to submit," said Mike Exton, Chief Executive Officer, Lexicon Pharmaceuticals, during the conference.
Understanding Sotagliflozin and SGLT Mechanisms
Sotagliflozin is a dual inhibitor of sodium-glucose cotransporter types 1 and 2 (SGLT1 and SGLT2). While SGLT2 inhibitors are widely established for treating type 2 diabetes and heart failure by promoting glucose excretion through the kidneys, SGLT1 inhibition specifically targets glucose absorption in the gastrointestinal tract.
This dual-action mechanism is designed to provide broader metabolic and cardiovascular benefits. In conditions like hypertrophic cardiomyopathy, where mechanical stress on the heart muscle impairs cardiac output, managing metabolic pathways represents an alternative therapeutic approach to traditional surgical or beta-blocker interventions.
Clinical Development and Market Stakes
The dual focus on hypertrophic cardiomyopathy and type 1 diabetes marks a critical phase for Lexicon Pharmaceuticals as the company navigates the high-cost environment of late-stage clinical trials. Successful readouts from the SONATA-HCM program and a cleared path for the type 1 diabetes resubmission will determine the commercial viability of sotagliflozin across multiple cardiology and endocrinology indications.
