Rein Therapeutics Resumes LTI-03 IPF Trial Following FDA Hold Removal
Rein Therapeutics has resumed its Phase II trial for IPF candidate LTI-03 after the FDA lifted a clinical hold, with funding secured through the first quarter of 2028.

Rein Therapeutics is moving forward with its Phase II clinical trial for LTI-03, a peptide-based therapy for idiopathic pulmonary fibrosis (IPF), after the resolution of a federal regulatory hold. The company reports a cash runway extending into the first quarter of 2028 and expects the resumption of testing to recover development timelines.
"LTI-03 is our drug for IPF, and the mechanism is pretty unique. This is a drug that mimics the protein caveolin-1. We're focused on caveolin-1 or CAV1, and CAV1 is a regulator of other proteins in the cell. We think of it as kind of an effector of homeostasis. It just promotes balance in the cell," said CEO Brian in a recent company presentation.
FDA Hold and Protocol Adjustments
The FDA lifted the clinical hold on the Phase II trial in September 2025. According to a company transcript, the regulatory clearance followed the submission of a briefing package. To satisfy safety requirements, the company adjusted its trial protocol to include enhanced lung function monitoring. Management estimates that the resolution of these concerns saved approximately 14 months of development time.
LTI-03 is administered via a twice-daily dry powder inhaler. The treatment mimics the CAV1 protein, which serves as a cellular homeostasis regulator. This approach differs from traditional antifibrotic therapies that focus on slowing the progression of lung scarring.
Financial Position and Market Context
Rein Therapeutics secured $57.5 million in financing in April 2024 to support its clinical pipeline. This capital provides the company with liquidity to sustain operations through Q1 2028, addressing immediate concerns regarding funding for the Phase II trial.
As of September 14, 2026, Rein Therapeutics (RNTX) stock was priced at $0.80. The company’s ability to maintain its development schedule is a focus for stakeholders, as the biotech sector faces scrutiny regarding the cost of clinical trials and the timeline for achieving regulatory milestones.
